Description
TB500, short for Thymosin Beta-4, is a synthetic peptide analog of the endogenous protein thymosin beta-4, which is naturally present in nearly all mammalian cells. Its molecular formula is C212H350N56O78S, with a molar mass of 4963.44 and CAS number 77591-33-4. It is a 43-amino-acid peptide that plays a key role in tissue repair and wound healing.
In scientific research, TB500 has been extensively studied for its biological activities. Preclinical in vitro studies have shown that TB500 (15-25 μg/ml) can promote cell migration and wound healing by enhancing ATP-induced intracellular calcium ion increase and activating purinergic P2 receptors. In vivo experiments on BALB/c mouse skin wound models have demonstrated that topical administration of TB500 (15 μg/30 μl, once daily for 10 days) significantly accelerates wound healing. Additionally, it has been found to be used as a doping agent in horse and greyhound racing due to its tissue repair properties.
The biological mechanism of TB500 mainly involves inhibiting the AKT signaling pathway and binding to actin, thereby exerting anti-fibrotic and wound-healing effects. It promotes endothelial cell differentiation, angiogenesis in dermal tissues, keratinocyte migration and collagen deposition, while reducing inflammation. Its active ingredient, Ac-Tβ4 17-23, also prevents apoptosis, promotes cell survival and immunomodulatory activity by stimulating T cell proliferation.
